What is Epithalon (Epitalon)? The AEDG tetrapeptide in published research

Reviewed by Floris, Vitality Peps Research TeamEditorial policy 8 min readPublished Updated

Epithalon is the synthetic tetrapeptide Ala-Glu-Asp-Gly (AEDG), designed from the amino acid composition of Epithalamin, a bovine pineal gland extract. In human fibroblast cultures it was reported to induce telomerase and lengthen telomeres, a finding a 2025 study repeated in normal human cells.

No controlled human trials. More than five controlled rodent studies (mainly from one St Petersburg group) on lifespan and spontaneous tumours, plus human and animal cell culture studies on telomerase.

Epithalon, also spelled Epitalon or Epithalone, is a four amino acid peptide that has been studied for more than 25 years, mostly in the context of ageing research. Much of the early literature comes from one group in St Petersburg, and newer work from other laboratories has revisited some of its claims in cell models. This guide sets out where Epithalon comes from, how its mechanism is described, what was observed in cells and animals, and where the evidence stops.

Where does Epithalon come from?

Epithalon traces back to Epithalamin, a peptide extract of bovine pineal glands studied in Russia from the 1970s onward. Researchers at the St Petersburg Institute of Bioregulation and Gerontology, led by Vladimir Khavinson, analysed the amino acid composition of that extract and synthesised a single tetrapeptide based on it: Ala-Glu-Asp-Gly, written AEDG. A 2025 review noted that the tetrapeptide was synthesised before it was detected in pineal gland polypeptide solution.

Epithalon is a short linear peptide with two acidic residues, glutamic acid and aspartic acid, which make it strongly negatively charged at neutral pH. Its calculated molecular weight is about 390 g/mol. The same 2025 review pointed out that although its biological effects have been studied for 25 years, physico-chemical and structural studies of the peptide remain limited.

How is it described at the molecular level?

No single receptor for Epithalon has been identified. Reviews describe several reported actions: an influence on melatonin synthesis in the pineal gland, changes in interleukin-2 mRNA, effects on thymus cell responses in mice, and increased activity of enzymes including acetylcholinesterase, butyrylcholinesterase and telomerase. The 2025 review concluded that it remains uncertain whether these are the only mechanisms.

The research group that developed it has proposed an epigenetic mechanism, in which the short peptide interacts with DNA or histones to change gene expression. A 2020 study reported that AEDG raised the expression of neuronal differentiation markers such as Nestin and Doublecortin by 1.6 to 1.8 times in human gingival mesenchymal stem cells, and molecular modelling suggested binding to histones H1/3 and H1/6. This proposed mechanism has not been confirmed by independent groups.

The telomerase finding is the best known. In 2003, Epithalon added to telomerase-negative human fetal fibroblasts induced expression of the catalytic subunit, telomerase activity and telomere elongation. A 2025 study from Brunel University London measured this in more detail and found telomere lengthening in normal epithelial and fibroblast cells through hTERT and telomerase, and in breast cancer lines through the alternative lengthening of telomeres pathway.

What has been observed in published research?

  • Lifespan and chromosomes (mouse): in female SHR mice given Epithalon monthly from three months of age, mean lifespan did not change, but maximum lifespan rose by 12.3 percent, bone marrow chromosome aberrations fell by 17.1 percent and leukaemia was less frequent.
  • Spontaneous mammary tumours (mouse): in HER-2/neu transgenic mice, Epithalon was associated with fewer and smaller mammary tumours and a 3.7 fold lower HER-2/neu mRNA level in tumours, compared with saline.
  • Pineal gland under stress (rat): after intranasal administration, a small rise in c-Fos protein was seen in pineal cells of stressed rats only, and stress-induced changes in pineal tissue were partly reduced.
  • Telomeres (human cells): telomerase activity and telomere elongation were reported in fetal fibroblasts in 2003 and in normal epithelial and fibroblast cells in 2025.
  • Oocytes (mouse and cow, in vitro): Epithalon in culture medium reduced reactive oxygen species and spindle defects in ageing mouse oocytes, and a 2025 study linked it to higher maturation rates of bovine oocytes.
  • Retinal cells (human cell line): in ARPE-19 cells exposed to high glucose, a 2025 study reported better wound closure and higher antioxidant gene expression with Epithalon.

What the evidence does not establish

Much of the animal literature on Epithalon was produced by one network of researchers, and several key studies were published in journals with limited independent replication. Lifespan results were modest and partly inconsistent: in the SHR mouse study, mean lifespan did not change at all. Rodent tumour findings do not translate directly to other species.

The telomerase findings come from cell culture. Lengthening telomeres in a dish is not the same as slowing ageing in an animal, and the 2025 finding of ALT activation in cancer cell lines adds a reason for caution. No randomised controlled human trials have been published, and a 2015 report found Epithalon in two illegally sold preparations, which illustrates why research material needs a documented identity and purity.

Which formats does Vitality Peps supply?

Vitality Peps supplies Epithalon as a 10 mg lyophilised vial for laboratory research, documented by a batch certificate of analysis that confirms identity and purity. Bacteriostatic water is listed separately for reconstitution.

How is it handled in the laboratory?

Lyophilised Epithalon is stored cold and protected from light. Before use the vial is brought to room temperature and reconstituted with bacteriostatic water added slowly down the side of the vial, then swirled gently. The working concentration is calculated from the actual mass on the batch certificate. Reconstituted solution is refrigerated, labelled and dated.

Research use only

Epithalon supplied by Vitality Peps is for in vitro laboratory research only. Not a medicine, not a supplement, not for human or veterinary consumption. Nothing above is instructions for use.

Frequently asked questions

Are Epithalon, Epitalon and AEDG the same compound?

Yes. Epithalon, Epitalon and Epithalone are spellings of the same tetrapeptide, and AEDG is its one letter amino acid code: alanine, glutamic acid, aspartic acid and glycine. It should not be confused with Epithalamin, the bovine pineal gland extract whose amino acid composition was used to design it. Epithalamin is a mixture; Epithalon is a single defined peptide.

What is the telomerase finding about?

Telomeres are protective caps at the ends of chromosomes that shorten as cells divide, and telomerase is the enzyme that can rebuild them. A 2003 study reported that Epithalon induced telomerase activity and telomere elongation in human fetal fibroblasts. A 2025 study found telomere lengthening in normal human cells through telomerase, and in breast cancer cell lines through a different route called ALT.

Has Epithalon been tested in human clinical trials?

No randomised, placebo-controlled trials of Epithalon were found in PubMed. Older Russian publications describe clinical observations with the related extract Epithalamin and with peptide bioregulators, but these were not designed as controlled trials. The published evidence for Epithalon itself is cell culture work and animal studies, mainly in mice and rats.

What did the animal lifespan studies show?

In a 2003 study of female SHR mice, Epithalon did not change food intake, body weight or mean lifespan. It was associated with a 12.3 percent longer maximum lifespan, fewer chromosome aberrations in bone marrow and less leukaemia, but no change in total tumour incidence. Most such studies came from one research group and have not been widely repeated.

Is the telomerase effect a concern in cancer research?

Researchers have raised this question, because many cancer cells keep dividing by maintaining their telomeres. The 2025 study found that Epithalon lengthened telomeres in two breast cancer cell lines through ALT activation. Older mouse studies reported fewer or smaller spontaneous mammary tumours. These findings point in different directions and have not been resolved.

How is Epithalon handled in the laboratory?

The lyophilised powder is kept cold and protected from light until use. It is reconstituted with bacteriostatic water at room temperature, and the working concentration is calculated from the actual mass on the batch certificate rather than the label alone. The solution is refrigerated, labelled and dated, as described in the storage and reconstitution guides.
